5-Amino-1MQ is a small-molecule quinolinium compound — not a peptide — that selectively inhibits nicotinamide N-methyltransferase (NNMT), a cytosolic enzyme involved in the methylation of nicotinamide using S-adenosylmethionine (SAM) as a methyl donor. NNMT is overexpressed in adipose tissue and certain metabolic contexts. By inhibiting NNMT, 5-Amino-1MQ has been shown to increase intracellular NAD+ and SAM availability, upregulate polyamine flux enzymes, and alter histone methylation patterns in preclinical adipocyte models.
